Doctoral Dissertation in Medical Sciences Reveals the Potential of Purple Sweet Potato Extract in Overcoming Obesity

Obesity is a major health problem throughout the world. Several studies confirm that consumption of foods rich in anthocyanins can prevent obesity in healthy subjects and can reduce body weight in obese subjects. This study aims to prove the effect and mechanism of purple sweet potato tuber extract on levels of malondialdehyde (MDA), TNF-α, leptin, phosphorylated AMPK (p-AMPK) in inhibiting the increase in body weight of male Wistar rats induced by a high-fat diet.


The design of this research is the randomized post test only control group design. A total of 30 male Wistar rats that met the eligibility criteria were randomized into 3 groups, namely treatment, control and normal. The treatment group was given a high-fat diet and 1 ml of 100 mg/ml purple sweet potato tuber extract for 12 weeks. The control group was given a high-fat diet and 1 ml of saline, while the normal group was given a standard diet and 1 ml of saline. MDA, TNF-α, leptin and p-AMPK levels were examined using enzyme-linked immunosorbent assay (ELISA). The data obtained were then subjected to descriptive and inferential statistical analysis, as well as path-analysis.


The results showed that the mean MDA level in the treatment group was lower than the control group (p=0.002). The mean TNF-α level in the treatment group was lower than the control (p= 0.038). The mean leptin level of the treatment group was lower than the control (p=0.002). The mean p-AMPK level in the treatment group was higher than the control (p=0.001). The mean body weight of the treatment group was lower than the control (p=0.024). Giving purple sweet potato tuber extract had a direct effect on reducing MDA levels (B: 0.79), TNF-α (B: 0.43) and leptin (B: 0.38), but had no direct effect on p-AMPK levels. as well as body weight. An inverse relationship was found between p-AMPK and levels of leptin (B: -0.44), TNF-α (B: -0.21) and MDA (B: -0.07). There was also a direct relationship between p-AMPK levels and weight loss (B: 0.06).


Thus, it can be concluded that purple sweet potato tuber extract has been proven to inhibit the increase in body weight of male Wistar rats induced by a high-fat diet through the mechanism of increasing p-AMPK levels due to decreasing levels of leptin, TNF-α and MDA.
