Doctoral Findings of Medical Science Prove the Potential of I-PRF to Reduce Inflammation and Increase Dental Pulp Regeneration

The potential of Injectable Platelet Rich Fibrin (I-PRF) in reducing pulp tissue inflammation and stimulating regeneration of dental stem cell tissue, where the preparation in liquid form is easily inserted into the root canal. PRF is rich in cytokines and growth factors that can increase cell proliferation and differentiation, increase angiogenesis, act as a matrix for tissue growth, regulate inflammatory reactions and anti-infection. This study aims to prove that the administration of I-PRF reduces TNF-a and IL-1b levels and increases the expression of DMP-1 and DSPP in the dental pulp of Wistar rats given lipopolysaccharide. The design of this study was a true experimental using a randomized posttest only control group design. The sample was 15 Wistar rats with 2 molar teeth each so that 30 Wistar rat teeth were used with a control group of 15 rat teeth and a treatment group of 15 rat teeth. The male rats used weighed 200-250 grams and were 4-5 months old. The teeth were cavities opened until the pulp was exposed, after which lipopolysaccharide was given to stimulate inflammation. The control group was not given any drugs, while the treatment group was given I-PRF. Then the rat dental pulp tissue was taken after the 7th day. The data observed by ELISA examination were TNF-a and IL-1b. RT qPCR was used to examine the relative expression of DMP-1 and DSPP by including beta actin to normalize sample variation. Data were analyzed using the independent t test to obtain differences in each variable. The results showed that TNF-a levels in the treatment group were 5.99 ± 2.47 pg / ml compared to the control group of 26.36 ± 2.21 pg / ml (p < 0.05) and IL-1b levels were 29.22 ± 4.79 pg / ml compared to the control group of 62.45 ± 5.81 pg / ml (p < 0.05). The results of the relative expression of DMP-1 and DSPP showed an increase (upregulated) of 2.22 and 3.24. From the results of the study, it can be concluded that the administration of Injectable Platelet Rich Fibrin to the dental pulp of Wistar rats previously given lipopolysaccharides reduces TNF-a and IL-1b levels and increases DMP-1 and DSPP expression.
