Doctoral Research in Medical Sciences Reveals the Potential of Purple Sweet Potato Extract as Antifibrotic Therapy in Kidney Disease

Uric acid is known to trigger oxidative stress, in vitroRE stress , epithelial to mesenchymal transition (EMT), cell death and renal fibrosis. Ethanol extract of purple sweet potato tubers (Ipomoea batatas L.) has antioxidant properties, but in vivo research on the antifibrotic effect of ethanol extract of purple sweet potato tubers on urate nephropathy models has never been done. This study aims to determine the effect of ethanol extract of purple sweet potato tubers on proapoptotic RE stress markers (C/EBP homologous protein or CHOP), EMT (N-cadherin expression), and myofibroblast expansion (α-SMA expression) in the kidneys of mice given exposure to uric acid.


In this study, the design used was a randomized post-test only control group design. Swiss strain mice (male, body weight 30-40 g, 22 mice) were divided into control groups given intraperitoneal uric acid (125 mg/kg BW) and distilled water orally for 7 days, and treatment groups given uric acid, and ethanol extract of purple sweet potato tubers (400 mg/kg BW) orally for 7 days. The extract was made by maceration method using 96% ethanol solvent. Quantification of CHOP levels was performed using ELISA, while N-cadherin and α-SMA expression was studied using immunohistochemical methods. Kidney preparations stained with hematoxylin-eosin (HE) and Masson's trichrome stain (MST) were made to support the results found. The results were analyzed using IBM SPSS software.


In the kidneys of mice exposed to uric acid, CHOP levels in the control and treatment groups were not significantly different. The absorbance of N-cadherin expression in the control group was significantly higher than the treatment group. The absorbance of α-SMA in the control group was significantly higher than that in the treatment group. Renal nephropathy and fibrosis were confirmed on HE and MST preparations, with qualitatively more severe damage found in the control group.


Administration of purple sweet potato tuber extract (Ipomoea batatas L.) at a dose of 400 mg/kgBB orally for 7 days produced renoprotective effects on the kidneys of mice treated with uric acid, through suppression of EMT and expansion of myofibroblasts.
