Premature Ovarian Insufficiency (POI) is a decline in ovarian function before the age of 40, which negatively impacts fertility. One emerging therapeutic innovation is intraovarian injection of Autologous Platelet-Rich Plasma (APRP), which has the potential to improve ovarian function. This study aims to demonstrate improved ovarian function through increased AMH, Estradiol, and Estrogen Receptor beta (ERβ) levels, as well as decreased FSH levels in Wistar rats with POI.


This study used a randomized, post-test-only control group design, involving 28 female Wistar rats with POI. The samples were divided into four groups: untreated control (K1), NaCl control (K2), APRP treatment with a dose of 0.25 ml (P1), and a dose of 0.375 ml (P2). AMH, FSH, and Estradiol levels were measured using ELISA, while ERβ expression was assessed through immunohistochemistry on day 14 post-injection. Data were analyzed using one-way ANOVA, Welch's ANOVA, and Kruskal-Wallis.


The results showed significant differences (p< 0.05) in all post-treatment parameters. A consistent upward trend was observed in hormonal parameters, with AMH levels reaching their highest peak in the P2 group (15.47±1.85) compared to K1 (6.11±0.93). Estradiol levels in P2 (1836.77±246.67) increased significantly, almost fourfold over the control. However, FSH levels showed a linear increase across all groups, with the highest value in P2 (4.42±0.28). Meanwhile, ERβ expression only showed a significant difference in the ratio of K1 to K2 and P2, while the ratio between P1 and P2 tended to be stable.


Intraovarian injection of APRP for 14 days improved ovarian function in Wistar rats with POI by improving the hormonal profile and estrogen receptor expression. Although improvements in the Hypothalamic-Pituitary-Ovarian (HHO) feedback mechanism have not yet been fully demonstrated, APRP demonstrates strong regenerative potential. Further research is needed to optimize dosage and longer observation durations for optimal results.
