Doctor of Medical Sciences Assesses the Potential of Purple Sweet Potato Tuber Extract as a Natural Solution to Overcome Postmenopausal Skin Aging

Menopause occurs in women aged between 45-55 years, causing a decrease in estrogen hormone levels, a decrease in physiological functions ranging from antioxidant capacity to triggering aging, especially skin aging referred to as Estrogen Deficient Skin (EDS) which is characterized by decreased skin collagen production. Efforts to overcome the problem of skin aging in EDS with hormone replacement therapy (TSH), but there is a risk of malignancy so that natural ingredients that have phytoestrogen properties as estrogen substitutes are given, one of which is purple sweet potato tuber which contains anthocyanins.


This study aims to prove that skin aging in the form of decreased collagen density in EDS can be overcome by giving ethanol extract of purple sweet potato tubers through the mechanism of higher ERβ and IGF-1 levels, and lower MMP-1 in Wistar rats after bilateral ovariectomy.


This experimental study used a randomized post test only control group design method, in the Histology and Biochemistry Division of the Integrated Biomedical Laboratory, Faculty of Medicine, Udayana University. Eighteen female Wistar rats that had been bilaterally ovariectomized were randomly divided into three groups, namely the control group (P0) which was given distilled water + Tween20, the treatment group of ethanol extract of purple sweet potato tubers + Tween20 at a dose of 400 mg / kgBB / day (P1) and a dose of 600 mg / kgBB / day (P2) orally for 3 months. On the 91st day, rat back skin tissue was taken with a size of 10 mm and a depth of 3-4 mm to measure the levels of ERβ, IGF-1, MMP-1 and collagen density. One Way ANOVA and Post Hoc LSD were used to test the differences in the mean levels of ERβ, IGF-1, MMP-1 and collagen density in the three groups followed by path analysis test.


The results showed that the mean levels of ERβ, IGF-1, MMP-1 and collagen density in skin tissue after treatment were significantly different. Mean ERβ levels were significant between P1 and P0 and P2 and P0, but not significant between P1 and P2, mean IGF-1 levels were significant between P1 and P0, P2 and P0, but not significant between P1 and P2, mean MMP-1 levels were significant between P1 and P0, P2 and P0 and P1 and P2, mean collagen density was significant between P1 and P0, P2 and P0 but not significant between P1 and P2. Path analysis found the effect of ERβ on collagen density was 56.1%, and significant with a value of p < 0.05, and the effect of ERβ, IGF-1 and MMP-1 on collagen density was 84%.


This study concluded that the administration of ethanol extract of purple sweet potato tubers at a dose of 400 mg / kgBB / day containing anthocyanins for 3 months was proven to increase the collagen density of Wistar rat skin tissue after bilateral ovariectomy through increased collagen synthesis with the mechanism of higher ERβ and IGF-1 levels, and inhibiting collagen degradation through lower MMP-1 levels in skin tissue.
