Doctoral Research in Medical Sciences Reveals the Effectiveness of Cemcem Leaf Extract in Healing Wounds in Hyperglycemic Rats

Located in the Postgraduate Hall, Fl. III Udayana University Postgraduate Building, the Doctoral Promotion exam took place with the promovenda candidate, Ns. Desak Made Widyanthari, S.Kep., M.Kep., Sp.Kep.MB., with the dissertation title "Study of the Molecular Pharmacological Activity of Topical Cemcem (Spondias pinnata (L.f) Kurz) Leaf Extract and Its Effect on the Expression of Superoxide Dismutase (SOD) , Nuclear Factor-Kappa Beta, Soluble IL-6 Receptor Levels and Wound Healing in Hyperglycemic Rats". (10/7/2024)

The wound healing process consists of several stages involving biochemical reactions and cellular responses. Hyperglycemia causes oxidative stress so that wound healing is not optimal. Cemcem leaves (Spondias pinnata (L.f) Kurz) have been proven to contain flavonoids which act as antioxidants and anti-inflammatories. This study aims to explain the molecular pharmacological mechanisms of the role of antioxidants and anti-inflammatories in the wound healing process in hyperglycemic mice after being given a topical gel preparation of cemcem (Spondias pinnata (L.f) Kurz) leaf extract.

The research was carried out at the Integrated Biomedical Laboratory, Faculty of Medicine, Udayana University from February to August 2023. This research was true experimental with a post test only control group design on 40 hyperglycemic mice which were randomly divided into five groups: negative control group, positive control group and The treatments given were 2.5%, 7.5% and 15% cemcem leaf gel extract. Cemcem leaf extract gel was applied twice a day for 14 days to the punch biopsy wound on the back of each mouse. SOD expression, NF-κB, sIL-6R levels and wound healing were observed on days 0, 3, 7 and 14. Statistical tests used one way ANOVA and Kruskal Wallis with a significance value of 0.05.

SOD expression was significantly higher in the treatment group compared to the control group (p=0.000). NF-κB expression was significantly lower in the treatment group compared to the control group (p=0.003). There was no difference in sIL-6R levels between the treatment and control groups (p>0.05). The mean wound diameter of the treatment group was smaller than the control group (p=0.000). The fastest wound healing was found in the group of mice given a 15% dose of cemcem leaf extract gel. Cemcem leaf extract gel has the potential effect of increasing wound healing in hyperglycemic mice.
